Study on the Phytochemical and Antioxidant Activity of Hypericum ascyron L.

Abstract

This study investigated the phytochemical composition and antioxidant activity of Hypericum ascyron L. collected in Mongolia. The aerial parts were extracted with 70% ethanol and successively partitioned with n-hexane, chloroform, ethyl acetate, and n-butanol. Phytochemical investigation of the fractions using column chromatography led to the isolation of seven compounds: kaempferol-3-O-glucoside (I), kaempferol-3-O-rutinoside (II), quercetin (III), isoquercetin (IV), chlorogenic acid (V), p-coumaric acid (VI), and hyperforin (VII). Their structures were elucidated by <sup>1</sup>H NMR and LC-MS/MS analyses and confirmed by comparison with published literature data. The antioxidant activity of the 70% ethanol extract was evaluated using the DPPH radical scavenging assay, yielding an IC<sub>50</sub> value of 75.78 ± 1.18 μg/mL. The total phenolic content, determined by the Folin–Ciocalteu method, was 4.71 ± 0.39% (w/w) of the extract. The isolation of flavonoids, phenolic acids, and hyperforin demonstrates that H. ascyron is a rich source of bioactive secondary metabolites. The observed antioxidant activity is likely attributable to its phenolic constituents, highlighting the potential of this Mongolian medicinal plant as a natural source of antioxidant compounds.

Их Алтан Цэгцүүхэй (Hypericum ascyron L.) ургамлын фитохими болон антиоксидант идэвхийн судалгаа

Хураангуй. Энэхүү судалгааны ажлын зорилго нь Монгол оронд ургадаг эмийн ургамал болох Их Алтан Цэгцүүхэй (Hypericum ascyron L.) ургамлын газрын дээд хэсгийн химийн найрлага болон антиоксидант идэвхийг тодорхойлоход оршино. Ургамлын дээжийг 70% этилийн спиртээр мацерацын аргаар хандалж гексан, хлороформ, этилацетат, н-бутанолын фракцуудад бүлэглэн хандлав. Этилацетат болон н-бутанолын фракцаас баганат хроматографийн аргаар кемпферол-3-O-глюкозид (I), кемпферол-3-O-рутинозид (II), кверцетин (III), изокверцетин (IV), хлорогенийн хүчил (V), р-кумарины хүчил (VI), гиперфорин (VII) зэрэг нийт 7 нэгдлийг химийн цэврээр ялган авч, тэдгээрийн бүтэц байгууламжийг 1Н ЦСР спектр болон шингэний хроматографи-масс спектрометрийн аргаар тодорхойлов. Ургамлын 70% этилийн спиртийн хандад антиоксидант идэвхийг 2,2-дифенил-1-пикрилгидразил (DPPH)-ын чөлөөт радикалыг саатуулах аргаар үнэлэхэд IC50 утга 75.78±1.18 мкг/мл бол нийт фенолт нэгдлийн агууламжийг Фолин-Чокалтеугийн аргаар тодорхойлоход 4.71±0.39 % байв.  Энэ нь H. ascyron ургамал нь флавоноид болон фенолт нэгдлээр баялаг бөгөөд антиоксидант идэвхтэй болохыг харуулж, Монгол оронд ургадаг энэхүү ургамлын фитохими болон антиоксидант шинж чанарын талаарх анхны мэдээллийг бүрдүүлж байна.
