Enzymatic activities of actinomycetes isolated from soil in Tuv province of Mongolia

Abstract

Actinomycetes found in diverse ecosystems around the world are characterized by wide range of clinically and industrially important natural products. In this study, agricultural soil samples were collected from Tuv province of Mongolia. Two actinomycetes strains (N23, N24) were isolated and selected for polyphasic approaches, including morphological, physiological and phylogenetic analysis. Morphological characteristics of strains were observed on different international streptomyces project media (ISP media). All strains were aerobic, gram-positive that have branches substrate mycelium with aerial mycelium differentiated. Phylogenetic analysis of these isolates revealed that they share highest similarity with Streptomyces shaanxiensis CCNWHQ 0031(T) FJ465151 (N23) and Streptomyces wuyuanensis FX61T(T) (JX010725) (N24), based on 16S rRNA gene sequencing. Then, enzymatic activities of actinomycetes were evaluated. The strain N23 possessed amylase and urease activities. The strain N24 showed good enzymatic activities with the enzyme amylase, urease, lipase, and protease activities, respectively.

Газар тариалангийн хөрснөөс ялгасан актиномицетийн ферментийн идэвхийн судалгаа

Хураангуй: Төв аймгийн нутаг дах газар тариалангийн хөрснөөс ялгасан N23, N24 дугаартай актиномицетийн өсгөврүүдэд филогенетикийн анализ хийж, амилаза, липаза, уреаза болон протеаза ферментийн идэвхийг тодорхойлов. Өсгөврүүдийн морфологийн шинж чанарыг ISP тэжээлт орчинд өсгөвөрлөж, субстрат мицелийн өнгө, агаарын мицелийн өнгө, спор үүсэлтийг тодорхойлов. 16S рРНХ генийн нуклеотидын дарааллыг тогтоож, филогенетикийн анализ хийхэд N23 өсгөвөр нь Streptomyces shaanxiensis CCNWHQ 0031(T) FJ465151 омогтой 100% төстэй учир Streptomyces shaanxiensis, N24 өсгөвөр нь Streptomyces wuyuanensis FX61T(T) (JX010725) омогтой 98.1% төстэй учир Streptomyces wuyuanensis төрөл зүйлд хамаарч байгааг тодорхойлов. N23 өсгөврийн ферментийн идэвхийг тодорхойлоход амилаза болон уреаза ферментийн өндөр идэвхтэй байна. N24 өсгөвөр нь амилаза, липаза, уреаза болон протеаза ферментийн өндөр идэвхтэй байгаа нь цаашид хүнс, хөдөө аж ахуй болон хөрсний нөхөн сэргээлтэнд ашиглах боломжтой нь харагдаж байна.
